Summary: According to the present opinions, the effect of free radicals on cell-contained organic compounds is one of the main etiological factors resulting in age-related macular degeneration development. Sources of oxygen free radicals, their interaction with lipids, proteins, carbohydrates and nucleic acids as well as effects of oxidation of the organic compounds were described. In particular, the oxidation of the polyunsaturated fatty acids contained in external photoreceptor segment membranes as well as the deposition of the reaction products (as lipofuscin) in retinal pigment epithelium cells were taken into consideration. The importance of the lipofuscin occuring phototoxic reactions and photosensitization of hemoglobin precursors in choriocapillaris for an oxidative damage of retinal pigment epithelium and photoreceptors was emphasized. The most important chemical compounds (glutathione, carotenoids) present in the antioxidative retina system and protecting the retinal nerve cells against the disadvageous action of oxygen-reactive species were also presented.
