Association of Oxidative DNA Damages and the Effectiveness of their Repair with the Risk of Primary Open Angle Glaucoma Development in the Polish Population

Summary: Purpose: Glaucoma is the ocular disorder, which is characterized by progressive degeneration of retinal ganglion cells and loss of visual field. The numerous studies suggest that oxidative stress may be associated with the risk of primary open angle glaucoma. Elevated level of oxidative DNA damage was found in the human trabecular meshwork among patients with primary open angle glaucoma as compared to controls. The purpose of our study was to evaluate the association between the level of oxidative DNA damage, repair capacity after hydrogen peroxide treatment and glaucoma pathogenesis.
Methods: Alkylation version of comet assay test was used to analyze level of endogenous damage and damage induced by hydrogen peroxide (10 µM and 20 µM), it the effectiveness of repair of oxidative DNA damage during 120 minutes incubation was also evaluated. The Fpg and NTH enzymes were utilized to determine the level of oxidative DNA damage.
Results: The results of presented study have shown the elevated level of endogenous oxidative DNA damage and DNA damage induced by hydrogen peroxide in group of patients with primary open angle glaucoma as compared to controls. Additionally, we have indicated the decreased efficiency of repair of oxidative DNA damage in glaucomatous patients.
Conclusion: Our study indicate that there is a significant elevated level of oxidative DNA damage and decreased efficiency of DNA repair in group of patients of primary open angle glaucoma as compared to controls. These results suggest that an impairment of the BER pathway may be one of the risk factors of the development of primary open angle glaucoma.
