Molecular Mechanism of Cataractogenesis

Summary: Cataract is the main cause of vision deterioration and vision loss. It is estimated that cataract causes reversible blindness in 17 mln. people around the world and that this number is likely to increase to 40 mln. by 2020. Age, diabetes, poor nutrition, lower antioxidant index and increased light exposure are commonly known as risk factors for cataract formation. With advancing age, the lens undergoes significant changes: yellowing of the lens protein, development of covalent bindings on the intra- and intermolecular level. The main two processes responsible for these changes are oxidation and glycation. Recent research suggest potential role of L-kynurenine aminotransferases (KAT) in cataractogenesis. Despite numerous studies on mechanisms of cataractogenesis, the pathogenesis of this process is not well known. In this paper, we present the most recent knowledge in pathophysiology of cataractogenesis and the main theories explaining processes of cataract formation.
